Does this Thule work with 2021 Lexus RX 450h with factory crossbars?

I may have a solution for you, but the Thule Hull-a-Port Aero # TH849000-97 can only be installed on square crossbars and the Hull-a-Port Aero # TH849000 is only designed to fit in the T-Slots found on aftermarket crossbars. Neither option is designed to fit on the factory crossbars on your 2021 Lexus RX 450h, but there are similar carriers that will work.
Since you have factory crossbars I recommend considering the Yakima JayLow Kayak Carrier w/ Tie-Downs - J-Style or Post Style - Folding - 1 or 2 Kayaks # Y04073, which will clamp to the factory crossbars on your RX 450h and give you a J-Style carrier for your boat. The JayLow also folds down similar to the Hull-a-Port Aero carrier and comes with tie down straps included, so it's going to give you the same style and features, just with a different brand.

Yakima factory crossbars on a CX-50,. Slid t-bolts down Yakima infill, measured 6 in. spacing for T-bolts marked it with a pencil on top of crossbars. Laid the Aero on top. I had play once tightening down with provided nut.Added 1/2 in. lock washer works perfect.
