If your 2020 Toyota Highlander has the factory roof rack with crossbars running left to right, then you are all set to install a cargo box. If you just have the factory flush rails running front to back, then you will need to add an aftermarket crossbar setup such as Thule WingBar # TH711300 along with Fit Kit # TH22RE and Feet # TH49SC.

When choosing a cargo box, the main thing you need to look at is the crossbar spread, which is the distance from the center of one crossbar to the center of the next crossbar. My research tells me that your roof rack will have a crossbar spread of about 28-30" but you can measure to be sure. You can look at the list I attached which shows compatible boxes ranging from 16 cubic feet to 18 cubic feet.
