My plan is to town 3000-4000 lb over long distances and will be using a weight distribution system. Is this hitch designed to be used with weight distribution systems?

Yes, the Curt hitch part # C13043 that you referenced for your 2016 Lexus GX 460 can be used with a weight distribution system as it was rated for use with them.
For the weight of your trailer the FastWay E2 kit part # FA92-00-0600 would be a really great option. It has a tongue weight range of 300-600 lbs and uses trunnion bars which are great for ground clearance. For a 2 inch hitch ball you'd then want the part # EQ91-00-6080.
