Didnt see any answers going back as far as my truck. Will this work on a 2001 GMC Sierra 2500 HD SLE extended cab.

For your 2001 GMC Sierra Pop and Lock does have a powered tailgate lock option but it's not the part number you mentioned as it's part # PAL8110.
I attached an install video for this kit on a similar 2005 Sierra as yours as well.
