Yes, although we haven't test fit the Malone RunWay # MPG2148 on a 2008 Toyota RAV4 as of yet (July 2018), we have confirmed with Malone that this will fit your vehicle with a spare tire.
This bike rack doesn't come with locks but if you want to add some security you can use the Thule Passive Security Strap # TH533 for the rack and the Cable Lock # e98891 for your bike(s).
You may also be interested in the Softride Soft Hook-and-Loop Tie Downs # SR54FR to help keep your front tire from potentially damaging another bike or making contact with your vehicle. If any of your bikes have an alternative frame that doesn't have a horizontal cross tube like a traditional men's bike then an adapter bar like part # UB01 is recommended. This will help keep your bike level and increase ground clearance.
