My 2010 F-350 SRW CC has a 6” lift and custom mount 5th wheel brackets, will this kit work for it?

Since there are no 6 inch spacers to use with the Timbren springs # TUR350 using them on a truck with a 6 inch lift like your 2010 Ford F-350 would render the springs useless. There are other options though. You could use Firestone Ride-Rite Air Helper Springs # F2550 if your custom 5th wheel brackets have the hole in the center of them (above the centerline of the factory bump stops). Otherwise that hole will have to be drilled which is generally not an accepted practice because it voids the warranty on the brackets.
With the Firestone air bags you would also need spacers to compensate for the lift. Now a 6 inch lift doesn't necessarily mean that the distance between the top of the axle and the bottom of the frame increased 6 inches. That means that 6 inch spacers # F2375 could be too tall. The next size down is a 4 inch # F2371.
Another option is a RoadActive Suspension kit # RAS3611 which may or may not work depending on the type of lift kit used on your truck. I have linked a video showing a typical setup that you can compare to your existing suspension to determine if this type of system will work.
