When installing the Derale Series 8000 Plate-Fin Transmission Cooler Kit # D13504 on your 2008 GMC Acadia SLT2 V6 there really is no "correct" way to orient the cooler as it can be mounted with the ports being up, down, left, or right as you mentioned.
I have attached the installation instructions for you to check out, but for the hose connections you will need to do the following:
1. Disconnect the return transmission cooler line from the radiator.
2. Take the supplied Insta-dapt Adapter fitting and install onto radiator using
the appropriate Male or Female end.
The Insta-dapt fitting has a 1/2”-20 Thread and fits most vehicles
with 5/16” transmission cooler lines. For vehicles that are not compatible
with the Insta-dapt Adapter, please reference the Radiator Adapter Fitting
3. Carefully route the looped hose now attached to the cooler to the Instadapt
Adapter now connected to the radiator.
4. Using the supplied Hose Clamp, cut hose and attach to the Insta-dapt
Adapter.
5. Route the remaining hose to the disconnected transmission line and cut hose to length.
6. Using the supplied Hose Clamp, attach and tighten.

I have attached an installation video on a 2009 GMC Yukon that you can use as a reference.
