Does this harness I bought operate backup lights on a trailer?

The Tow Ready T-One Vehicle Wiring Harness you referred to, part # 118640 will feed the trailer running lamps, brake lamps and right and left directional signals, but not reverse lamps.
We do not have a plug and play wiring harness option that would provide a 5-pole, but you could hardwire one using part # 119177KIT. You would use the tester included in the kit to test the wires feeding the factory taillight assemblies to locate which wire carries the right turn, left turn, brake lamp, reverse lamp and running lights.
When you locate the right turn circuit, tap the green wire into it. Yellow wire goes to the left turn, red wire to brake lamp, blue wire to reverse lamps and brown wire to running lights. The white wire is grounded and the black wire would be extended and ran to the positive battery terminal, using the fuse holder included with the kit.
