Question:
Does the mirror have to be removed to tighten the adjusting screw or can it be done on the truck?

You should be able to adjust the tightness of the towing mirrors # KS81800 with them installed. However, the adjustment screw is accessed from the bottom of the mirror so it would be much easier to adjust with them off of the truck. Since taking them off is so easy, I recommend removing them to tighten up the mirrors. I have linked a video showing a typical installation.
