Question:

Does the foam washer go under the flange of the drain tube inside the sink, or above the flange thread-on ring on the under side of the sink? Also, where and how do I apply sealant to the sink and drain tube?

The foam washer that comes with # 34465SPR1303 sits underneath the stainless steel drain flange inside the sink. When you tighten the nut down on the other side it will compress the foam washer creating a seal. This should be enough to keep anything from leaking. Since water can only go down the drain you won't need any sealant where it connects to your existing plumbing either.
