Question:
Is the dimension shown the opening in the wall for the vent? The one I have has an overall dimension of 12 X 51/2inch outside of flange to outside of flange.

The Ventline Exterior Wall Vent # V2111-13 that you referenced features overall dimensions of 11-5/8 inches x 5-5/16 inches. The listed 10-1/2 inch x 3-3/4 inch measurements refer to the rough opening it will install in.
I have attached an dimensional diagram with more specific measurements labeled that you can check out as well.
