Question:
I just installed the controller and set up the level switch, no problem. When I tried to adjust the gain none of the lights will light. I hand activated the slide switch, pushed the brake pedal, adjusted the dial up and down but no lights? Does the trailer need to be hooked up to the truck to activate?

According to my information the Sentinel Brake Controller does have to be connected to a trailer to operate. There will be a green light that comes on when the controller identifies that the trailer is connected to the truck. After you have this then you will be able to set the gain.
If you continue to have no power to your controller after you connect the two, then it sounds like you are missing the 12 volt power wire being routed from your brake controller to the battery used in combo with a 30 amp circuit breaker # 38630, so that you do not overload the electrical system. You can run this wire and breaker with the use of # 10-1-1 and ring terminals like # DW05705-1.
