Yes, it's super common to replace a 3-hole faucet with a single, like the Empire Faucets RV Kitchen Faucet w/ Pull-Down Spout # EM42HR. The deck plate will allow you to cover the two inlet shank holes and as you tighten the middle inlet shank down it will prevent the plate from sliding around and exposing the holes. You'll just want to ensure that your middle inlet shank hole is wide enough to allow the 1-1/4" inlet shank of this faucet to sit inside of it.
