Will the 12inch wheels with a -0.5inch offset have enough clearance to fit over a 10inch brake drum on a 3500lb axle?

Your typical 3,500 lb hub and drum assembly like the # AKHD-545-35-EZ-K will only work with 13"-15" wheels. Since the # AM20149 are 12" wheels they will not fit, regardless of any offset. You will need a larger wheel to fit the hub and drum assembly.
Although we do not have a wheel with the same offset that would work, you could use the Dexstar Steel Mini Mod Trailer Wheel - 13" x 4-1/2" Rim - 5 on 4-1/2 # AM20253. This has the same bolt pattern with an offset of 0. Most trailer wheels have an offset of 0 so it is unlikely it would be a problem unless you have a very specific need for it.
