Will the factory rail covers need to be removed or drilled to mount the A4001666? Also wondering if it will be compatible with my tonneau cover.

The Adarac Pro Series Custom Truck Bed Ladder Rack # A4001666 mounts to the rails of your 2019 Tundra using the stack pockets and doesn't require any drilling to install. You may need to remove your stake pocket covers, but it will install over the rail caps without removal.
The Adarac Pro Series will be compatible with tonneau covers that mount inside the bedrails, but not those that mount on top of the bed rails. If you've got a tonneau cover that mounts on top of the bed rails there is not a ladder rack available that is compatible with it so you'd need to either remove the tonneau completely or go with a different one.
