Is Pop and Lock Powered Tailgate Lock PAL8500 Designed to Fit With or Without Manual Tailgate Lock

I have a 03 tundra and my tailgate has a lock on it know but I would like to be able to use Amy remote instead of key. Will this work for me

Yes, I spoke with Pop and Lock and the # PAL8500 which fits your 2003 Toyota Tundra fits the tailgate whether it came with a factory manual lock or not.
