In the picture of the closed position from behind, there appears to be a decent gap around the closure. Does this actually make a seal or is there air exchange even in the closed position?

The Valterra Heating and A/C Vent Register part # A10-3350VP like most vent covers is not completely air tight. There will still be a bit of air that can escape through the vent while closed. This is very common for duct work either in a home or RV.
If the above vent is not as air tight as you would like then you may want to consider a different style, the Valterra Heating and A/C Vent Register part # A10-3349VP which has an off white color but will form a better seal. I have attached a review video for you.
