For a good soft roll up Tonneau cover that will fit your 2016 Ford F-150 5-1/2 foot bed without trac system I recommend the BAK Revolver X2 # BAK39329. It is actually a combination vinyl and aluminum cover but it still rolls up. This design is also going to prevent the cover from flapping or bubbling.
To lock it you would just have to lock the tailgate because the cover cannot be rolled up without lowering the tailgate to access to the latches.
For a similar cover that doesn't have the aluminum you could use # TX597701. To lock it again you would have to lock the truck tailgate.
If you are interested in a retractable hard cover that has its own lock you can use the Retrax cover # RTT-80373 or # RT60373. It can be locked in any position and not just when closed. I have linked some videos of these covers for you.
