Hello, Im working on assembling a RV rig for full time, cross country travel. The least expensive way for me to go would be to use the 2008 E250 I already own as a tow vehicle excellent condition, 98k. Its towing capacity is 5,900 lbs. There are a number of lightweight trailers I could purchase with a GVW max of 3,500 lbs. Would it be wise to do this? Is it reasonable to expect this van to be able to work this hard, full time? I would greatly appreciate your honest, expert opinion.

The full size Ford vans, especially the 3/4 ton models like your E-250, are made to carry around a lot of weight. And since its towing capacity is 5,900 pounds, towing around a 3,500 pound trailer all the time would not be too much for the vehicle to handle.
However, there are some things you can do to make it easier for the van. You can add a transmission cooler because towing will put additional strain on the vehicle transmission. The most popular cooler option is # D13503.
Another easy upgrade is suspension enhancement. Adding helper springs such as # TFER35092B will take the strain off of the vehicle suspension and improve ride quality. After installing these helper springs you will not have to worry about them again.
With those 2 upgrades and the weight of the trailer well under the capacity of the van you would be in good shape!
