I am looking at using the Yakima stage two bike rack with the Roadmaster spare tire carrier rm-195125 on a Class Motor home 2024 Forest River 2350LE? My tire size is 225/75/16. Will This work with a spare tire mounted?

Great question, David!
The Yakima StageTwo Bike Rack # Y47VR requires at least 8-1/2" from the center of the hitch pin hole to the furthest part of the spare tire in order for it to fit properly. The center of the hitch pin hole on the Roadmaster Spare Tire Carrier # RM-195125 is about 5-5/8" behind the face of the mounting plate for your spare tire which means your tire can't sit more than 2-7/8" out from the mounting plate in order for you to be able to use the StageTwo.
For an alternative option that offers an additional 1-1/2" of clearance I recommend the Swagman Escapee # S39SR. This has the same basic features as the StageTwo but with this rack the tire can sit up to 4-3/8" out from the face of the Roadmaster mounting plate which is much more feasible.
