You would need to use a circuit tester like # 3808 if needed to determine the wire functions on the tail lights of your 2012 Ford Escape. This would only be if you are hardwiring in a harness. All of the wiring harnesses for the vehicle are T-Connectors that just plug in line with the vehicle wiring, see link.

The wiring harness that you have referenced, # 118251, is designed for vehicles that came with the factory tow package. A dealership can run the VIN to tell you how the vehicle is equipped and what options it has, included whether or not it has a tow package.

On a 4-Way trailer connector, white is ground, yellow is left turn and brake lights, green is right turn and brake lights, and brown is the running lights.
