Question:
Have a 2012 Toyota Sienna with Factory Crossbars. Would like to know if this Thule Tandem carrier TH558P will work with this vehicle, and the factory crossbars. If not, what is required to make it compatible. Also, if Thule crossbars are required, do the factory crossbars have to be removed?

The Thule Tandem Roof Mounted Bike Rack # TH558P that you referenced is only confirmed to fit standard Thule and Yakima bars, it would not fit on the factory cross bars of your 2012 Toyota Sienna.
You would first need a roof rack, I would get the Thule Roof Rack # THLB58 and # TH450. Once installed you would be able to put the # TH558P on your roof. I attached a video of this product on a very similar Toyota Sienna for you to check out also.
You will not need to remove your factory cross bars to install the Thule rack either.
