Question:
1. Would this cooler fit in our 2004 V6 Sienna and be appropriate for towing a tent camper weighting 2,000 pounds? 2. Where is the transmission line to cut for the installation? 3. Do you know of any special instructions for this type of vehicle?

Transmission coolers are not vehicle specific, so you can use the Derale Cooler, # D13501. It would be a good choice for what you plan on towing. Your vehicle should not require any special fittings or installation instructions beyond the basic installation method. I have included a link to a video showing a typical installation for you to view.
The recommended install method is to connect the return line from the factory cooler to the Derale cooler, then run the out from the Derale cooler back to the transmission. Depending on where you can mount the cooler will determine where you will need to cut the lines.
